Description

MOTS-c

Also known as: Mitochondrial ORF of the 12S rRNA type-c

MOTS-c is a peptide encoded by mitochondrial DNA, part of a group known as mitochondrial-derived peptides (MDPs), and is being studied for its potential impact on metabolic function, energy balance, physical performance, insulin sensitivity, and aging.
Frequently referred to as an “exercise-mimetic,” MOTS-c is of interest for its ability to modulate cellular metabolism, trigger stress-response mechanisms, and support cellular adaptation during metabolic stress.
Because of its links to mitochondrial activity and age-related metabolic changes, it has gained significant attention in longevity research and biohacking circles.
Key areas of scientific interest include:
– Enhancing metabolic efficiency and fat utilization
– Improving insulin sensitivity and blood glucose control
– Boosting exercise capacity and endurance
– Supporting mitochondrial function and energy generation
– Influencing pathways associated with longevity and healthy aging
Status: Currently in the preclinical phase, with early human trials starting. Classified as FDA Category 2, though a reclassification to Category 1 is expected following the April 15, 2026 HHS announcement. The change remains pending formal FDA rulemaking; a PCAC review is scheduled for July 23–24, 2026. Current evidence is based on animal studies.
Typical administration: 5–10 mg per week, delivered via subcutaneous injection, usually given 1 to 3 times weekly.
Primary research focus: Metabolic health, enhancement of exercise performance, and improvement of insulin sensitivity.
